چکیده

background and objectives: loop-mediated isothermal amplification is a novel nucleic acid amplification assay providing as a simple diagnostic tool for rapid identification of microbial diseases in developing countries. in this study, a lamp assay was established for yersinia enterocolitica , a leading cause of acute enterocolitis in young children. materials and methods: lamp assay was established with four primers targeting a specific locus for the detection of y. enterocolitica . the assay was conducted at 65°c in thermo block for 90min. the sensitivity of lamp was evaluated in com- parison to conventional pcr using ptz57r containing the target locus. finally, specificity was assessed using dna from common enteropathogenic bacteria. results: results showed that the sensitivity of lamp assay was 44-copy number, which was 10-fold higher than that of pcr. no cross-reactivity was observed when testing against other enteropathogenic pathogens. conclusion: this study showed that lamp assay is an alternative molecular diagnostic tool for infections with y. enteroco- litica . in addition, this method may be useful in diagnosis at field or in laboratories without pcr machine.

Rapid detection of infectious bursal disease by loop-mediated isothermal amplification for field analysis

Infectious bursal disease (IBD) is an immunosuppressive, acute and highly contagious illness of growing-poultry stock infected with infectious bursal disease virus (IBDV). It is common in Pakistan, causing potential economic losses throughout the year. The objective of the study is to propose a rapid, sensitive and specific diagnostic tool, and compare it with existing commonly used reverse tra...

A rapid loop-mediated isothermal amplification assay targeting hspX for the detection of Mycobacterium tuberculosis complex.

A rapid, simple, and low-cost diagnostic tool for tuberculosis (TB) detection is urgently needed in countries with a high TB burden. Here, we report a novel loop-mediated isothermal amplification (LAMP) assay targeting the hspX gene for the rapid detection of Mycobacterium tuberculosis, M. bovis, M. africanum, and M. microti. The specificity of this assay was evaluated using 4 reference str...

Development of a loop-mediated isothermal amplification assay for rapid detection of capripoxviruses.

Sheep pox (SP), goat pox (GP), and lumpy skin disease (LSD), caused by capripoxviruses (CaPVs), are economically important diseases of sheep, goats, and cattle, respectively. Here, we report the development of a loop-mediated isothermal amplification (LAMP) assay for rapid detection of CaPVs. LAMP primers were designed to target a conserved gene encoding the poly(A) polymerase small subunit (VP...

A reverse transcriptase-loop mediated isothermal amplification assay (RT-LAMP) for rapid detection of bovine viral diarrhea virus 1 and 2

Bovine viral diarrhea virus (BVDV) is a pathogen that infects cattle, and is globally important. It causes substantial financial losses to the livestock industry. In the current study, a one-step reverse transcriptase-loop-mediated isothermal amplification (RT-LAMP) assay was set up for rapid and efficient detection of BVDV. For this purpose, four primers were designed to recognize six distinct...
